Why boronic acids remain the workhorse coupling partner in medicinal chemistry.
Boronic acids are the most widely used nucleophilic partner in the Suzuki-Miyaura cross-coupling. Their stability, low toxicity and tolerance of a wide range of functional groups make them well suited to medicinal chemistry libraries and to scale-up. Storage under dry conditions and control of boroxine content are the practical considerations that matter most in manufacturing.
